RESUMO

This study is aimed at exploring the potential role of GSDMC in kidney renal clear cell carcinoma (KIRC). We analyzed the expression of GSDMC in 33 types of cancers in TCGA database. The results showed that the expression of GSDMC was upregulated in most cancers. We found a significant association between high expression of GSDMC and shortened patient overall survival, progression-free survival, and disease-specific survival. In vitro experiments have shown that the expression of GSDMC was significantly elevated in KIRC cell lines. Moreover, decreased expression of GSDMC was significantly associated with decreased cell proliferation. In summary, we believe that this study provides valuable data supporting future clinical treatment.

UCEC is one of the three common malignant tumors of the female reproductive tract. According to reports, the cure rate of early UCEC can reach 95%. Therefore, the development of prognostic markers will help UCEC patients to find the disease earlier and develop treatment earlier. The ALDH family was first discovered to be the essential gene of the ethanol metabolism pathway in the body. Recent studies have shown that ALDH can participate in the regulation of cancer. In our research, we explored the expression of the ALDH family in 33 cancers. It was found that ALDH2 was abnormally expressed in UCEC. Besides, in vivo and in vitro experiments were conducted to explore the effect of ALDH2 expression on the proliferation of UCEC cell lines. Meanwhile, the change of its expression is not due to gene mutations, but is regulated by miR-135-3p. At the same time, the impact of ALDH2 changes on the survival of UCEC patients is deeply discussed. Finally, a nomogram for predicting survival was constructed, with a C-index of 0.798 and AUC of 0.764. This study suggests that ALDH2 may play a crucial role in UCEC progression and has the potential as a prognostic biomarker of UCEC.

OBJECTIVE: The objective is to explore the effects of enhancing the activity of yeast ferulic acid decarboxylase (FDC1) on the level of 4-vinylguaiacol (4-VG) and the consumption of its precursor ferulic acid (FA) in top-fermented wheat beer. RESULTS: Expression of Bacillus pumilus FDC1 in brewer's yeast showed a better effect on the FDC1 activity than overexpression of the endogenous enzyme. The 4-VG content was increased by 34%, and the consumption time of FA was shortened from 48 to 12 h. Since the intracellular accumulation of the FDC1 substrate did not increase over time, to reduce the FA transport burden on cells and shorten the decarboxylation time, B. pumilus FDC1 was further secreted extracellularly. The resulted strain showed a 65% increase in 4-VG content in the FA-containing medium, and produced about 3 mg L-1 4-VG in the top-fermented wheat beer, increasing by 61% than control. However, further increasing the secretory expression level of FDC1 only accelerated FA consumption. CONCLUSIONS: These results suggested that appropriate secretion of bacterial FDC1 into wort could be used as a potential alternative strategy to increase the level of 4-VG in top-fermented wheat beer.

Microalgae harvesting is a major hindrance for the development of the microalgae industry. In this paper, short microwave treatment was used to assist the flocculation of Chlorella vulgaris with three flocculants, Fe3+ (FeCl3), chitosan, and Ca2+ (CaCl2). A microwave irradiation time of 20 s, and a pH of 10 was found to be the optimum condition. The harvesting efficiency could be significantly increased by 43.2%, 49.5% and 39.6%, respectively for Fe3+, chitosan, and Ca2+ assisted by microwave under these conditions. Microwave treatment did not cause any damage to the algal cells, and had no obvious influence on the lipid extraction. Microwave treatment decreased the concentration of the flocculants in culture medium after flocculation; this treatment enabled the reuse of the supernatant. This study provides a new and promising method of improving the flocculation efficiency for microalgae harvesting, by using microwave energy.

The interaction of 100-fs laser pulses with solid targets at laser intensities 10(16)-10(18)W/cm(2) has been investigated experimentally by simultaneous measurements of terahertz (THz) and second harmonic signals. THz yield at the front side of the target, which rises from the self-organized transient electron currents along the target surface, is found scaling linearly with the laser intensity basically. Measurements of specularly reflected light spectrum show clear evidence of resonance absorption. The positive effects of resonance absorption on surface current and THz radiation generation have been confirmed by two-dimensional (2D) particle-in-cell (PIC) simulations and angular-dependent experiments, respectively.

This work aimed at determining 4-vinylguaiacol and 4-vinylphenol in the top-fermented wheat beers using different wavelength and the mobile phase for HPLC. Best results for isocratic elution were obtained at 260 nm and the mobile phase comprising methanol/ultrapure water/phosphoric acid (400/590/10, V/V). Under these conditions, the retention time of 4-vinylguaiacol and 4-vinylphenol was 25 and 27min, respectively.

Surfactin has been known to inhibit proliferation and induce apoptosis in cancer cells. However, the molecular mechanisms involved in surfactin-induced apoptosis remain poorly understood. The present study was undertaken to elucidate the underlying network of signaling events in surfactin-induced apoptosis of human breast cancer MCF-7 cells. In this study, surfactin caused reactive oxygen species (ROS) generation and the surfactin-induced cell death was prevented by antioxidants N-acetylcysteine (NAC) and catalase, suggesting involvement of ROS generation in surfactin-induced cell death. Surfactin induced a sustained activation of the phosphorylation of ERK1/2 and JNK, but not p38. Moreover, surfactin-induced cell death was reversed by PD98059 (an inhibitor of ERK1/2) and SP600125 (an inhibitor of JNK), but not by SB203580 (an inhibitor of p38). However, the phosphorylation of JNK rather than ERK1/2 activation by surfactin was blocked by NAC/catalase. These results suggest that the action of surfactin on MCF-7 cells was via ERK1/2 and JNK, but not via p38, and the ERK1/2 and JNK activation induce apoptosis through two independent signaling mechanisms. Surfactin triggered the mitochondrial/caspase apoptotic pathway indicated by enhanced Bax-to-Bcl-2 expression ratio, loss of mitochondrial membrane potential, cytochrome c release, and caspase cascade reaction. The NAC and SP600125 blocked these events induced by surfactin. Moreover, the general caspase inhibitor z-VAD-FMK inhibited the caspase-6 activity and exerted the protective effect against the surfactin-induced cell death. Taken together, these findings suggest that the surfactin induces apoptosis through a ROS/JNK-mediated mitochondrial/caspase pathway.
